First line 5x+y =  -1

Subtract 5x from each side.

5x+y-5x = -1-5x

y = -1-5x

y = -5x-1

Slope of the first line m1 =-5.

Second line x-5y = 10

subtract x from each side.

x-5y-x = 10-x

-5y = -x+10

Divide by negitive 5 to each side.

-5y/-5 = -x+10/-5

y = -x/-5+10/-5

y = (1/5)x-2

Slope of the second line m2 = 1/5.

m1*m2 = -5*1/5 = -1

so the given lines are perpendicular lines.
